This research was conducted with the aim of investigating and identifying the effects of heavy metals Ni and Pb on the physiology of Asian sea bass with emphasis on enzyme levels and measurement of heavy metals (Ni, Pb) in the muscle tissue of sea bass farmed in cages. This research was conducted in the October of 2022, in Bushehr province, Kangan region. The samples after fixation with alcohol, were transferred to the laboratory and biometric indicators including weight and total length were measured. Then, a sample was prepared from the muscle tissues of each piece of fish and the concentration of heavy metals was obtained by Flame Atomic Absorption Spectrophotometry (FAAS). For study of tissue pathology, the muscle tissue sample was separated and the tissue passage steps, including dehydration, clarification and staining by the tissue passage device or histokinet, were done under a scheduled and stained by the H&E method, and the percentage of effect each heavy metal was analyzed separately. Aminotransferase, alkaline phosphatase and alanine aminotransferase enzymes were also measured using commercial kits of Pars Azmoun Company and according to the kit instructions. The results showed that the average concentration of nickel and lead metals in muscle tissue was 1.3 and 0.8 PPM, respectively, which is higher than WHO and FAO standards. The average concentration of alkaline phosphatase enzymes was 37.66, alanine amino transferase 75.16 and aminotransferase 47.16 U/L. In the investigation of the effect of heavy metals on the muscle tissue of Asian sea bass, complications such as amyloid cells, melanoma macrophase, destruction of muscle fibers and muscle atrophy were observed.
