One of the causes of widespread mortality in shrimp on farms is white spot virus that leads rapid and high death within a few days. Early detection prevents huge financial loss. The only method for early and reliable diagnosis of this virus is a molecular method. VP28 and protein kinase-related regions in virus genome, and also ribosomal 18S region as internal control from the shrimp genome are used for development a complex compatible PCR primers. extracted DNA from 200 samples of suspected shrimp were evaluated for detection of virus with standard kit IQ2000(Taiwan) and optimized multiplex PCR. Of all samples, 94 and 106 were positive (infected with the virus) and negative, respectively. Two samples were not concordance with reference kit, so , the sensitivity and specificity of optimized method were 98 and 100%, respectively. Based on the results, the optimized one-step method in this study can be used as standalone or complement method with reference kit for diagnosis of white spot virus.
